No. Fuel, working-time and other costs arise over all 400 km, even though no goods are carried on the empty 100 km leg.

Fuel consumption need not be the same for loaded and empty trucks; calculate actual costs using appropriate data.
A simple explanation
Variable costs depend on the amount of activity, and empty running does not reduce them to zero. A return load can improve vehicle utilisation if it fits the route, timing, equipment and legal requirements.
An empty truck still costs money.
